After reviewing the concept of the specific and non specific thalamo-cortical systems, the connectivity of the relay and intralaminar nuclei is analyzed as well as the recent data concerning the chemical identity of thalamic neurones, the concept and distribution of \"matrix\" and \"core\" neurones and its functional role. The intrinsic electrical properties of thalamic neurones, its mode of discharge--depending of the membrane potential level--and its functional significance in the context of the brain\'s global activity are discussed. Of special interest are the studies on the effects of lesion of the relay and intralaminar nuclei as well as its repercussion in the interpretation of the sensory perception. In this review, I discuss these mechanisms both in the adult and developing brain, relying on studies in which chloride and GABAergic currents were measured by electrophysiological and imaging techniques. The method provides not only the identification of the aforementioned hemoglobin and variants but also an accurate quantitation of their concentrations, particularly Hb F and A2, which are useful for the diagnosis of HPFH and beta-thalassemia, respectively.CONCLUSIONS: The simplicity of the sample preparation, superior resolution of the method, and accurate quantitation of hemoglobin concentration, combined with complete automation, make this an ideal methodology for the routine diagnosis of hemoglobin disorders in a clinical laboratory..
